Fuel prices are on the rise throughout the country, prompting West, a Republican from Florida?s 22nd district, to take to the virtual pages of Facebook to voice his displeasure with President Obama?s energy policies. Although most Americans likely share West?s concerns about rising fuel prices, the Congressman lost all sympathy when he mentioned what kind of vehicle he drives.

Nanoscale magnetic resonance imaging, quantum computer get nudge from new research

ScienceDaily (Feb. 23, 2012) ? Magnetic resonance imaging (MRI) on the nanoscale and the ever-elusive quantum computer are among the advancements edging closer toward the realm of possibility, and a new study co-authored by a UC Santa Barbara researcher may give both an extra nudge.

The findings recently appeared in Science Express, an online version of the journal Science.

Ania Bleszynski Jayich, an assistant professor of physics who joined the UCSB faculty in 2010, spent a year at Harvard working on an experiment that coupled nitrogen-vacancy centers in diamond to nanomechanical resonators. That project is the basis for the new paper, "Coherent sensing of a mechanical resonator with a single spin qubit."

A nitrogen-vacancy (NV) center is a specific defect in diamond that exhibits a quantum magnetic behavior known as spin. When a single spin in diamond is coupled with a magnetic mechanical resonator -- a device used to generate or select specific frequencies -- it points toward the potential for a new nanoscale sensing technique with implications for biology and technology, Jayich explained.

Among those possible future applications of such a technique is magnetic resonance imaging on a scale small enough to image the structure of proteins -- an as-yet unaccomplished feat that Jayich called "one of the holy grails of structural biology."

"The same physics that will allow the NV center to detect the magnetic field of the resonator, hopefully, will allow MRI on the nanoscale," Jayich said. "It could make MRI more accurate, and able to see more. It's like having a camera with eight megapixels versus one with two megapixels and taking a picture of someone's face. You can't see features that are smaller than the size of a pixel. So do they have three freckles, or do they all look like one big freckle?

"That's the idea," Jayich continued. "To resolve individual freckles, so to speak, to see what a protein is made up of. What we found in this paper suggests that it is possible, although a significant amount of work still needs to be done."

Though further into the future based on the approach used for this paper, Jayich said, there is also the potential for such a coupling to be advanced and exploited as a possible route toward the development of a hybrid quantum system, or quantum computer.

Jayich collaborated on the project with researchers Shimon Kolkowitz, Quirin Unterreithmeier, Steven Bennett, and Mikhail Lukin, all from Harvard; Peter Rabl, from the Institute for Quantum Optics and Quantum Information of the Austrian Academy of Science; and J.G.E. Harris, from Yale. The work was supported in part by the National Science Foundation, the Center for Ultracold Atoms, and the Packard Foundation.

USB stick can sequence DNA in seconds

It may look like an ordinary USB memory stick, but a little gadget that can sequence DNA while plugged into your laptop could have far-reaching effects on medicine and genetic research.

The UK firm Oxford Nanopore built the device, called MinION, and claims it can sequence simple genomes ? like those of some viruses and bacteria ? in a matter of seconds. More complex genomes would take longer, but MinION could also be useful for obtaining quick results in sequencing DNA from cells in a biopsy to look for cancer, for example, or to determine the genetic identity of bone fragments at an archaeological dig.

The company demonstrated today at the Advances in Genome Biology and Technology (AGBT) conference in Marco Island, Florida, that MinION has sequenced a simple virus called Phi X, which contains 5000 genetic base pairs.

Proof of principle

This is merely a proof of principle ? "Phi X was the first DNA genome to be sequenced ever," says Nick Loman, a bioinformatician at the Pallen research group at the University of Birmingham, UK, and author of the blog Pathogens: Genes and Genomes. But it shows for the first time that this technology works, he says. "If you can sequence this genome you should be able to sequence larger genomes."

Oxford Nanopore is also building a larger device, GridION, for lab use. Both GridION and MinION operate using the same technology: DNA is added to a solution containing enzymes that bind to the end of each strand. When a current is applied across the solution these enzymes and DNA are drawn to hundreds of wells in a membrane at the bottom of the solution, each just 10 micrometres in diameter.

Within each well is a modified version of the protein alpha hemolysin (AHL), which has a hollow tube just 10 nanometres wide at its core. As the DNA is drawn to the pore the enzyme attaches itself to the AHL and begins to unzip the DNA, threading one strand of the double helix through the pore. The unique electrical characteristics of each base disrupt the current flowing through each pore, enough to determine which of the four bases is passing through it. Each disruption is read by the device, like a tickertape reader.

Long strands, and simple

This approach has two key advantages over other sequencing techniques: first, the DNA does not need to be amplified - a time-consuming process that replicates the DNA in a sample to make it abundant enough to make a reliable measurement.

Second, the devices can sequence DNA strands as long as 10,000 bases continuously, whereas most other techniques require the DNA to be sheared into smaller fragments of at most a few hundred bases. This means that once they have been read they have to be painstakingly reassembled by software like pieces of a jigsaw. "We just read the entire thing in one go," as with Phi X, says Clive Brown, Oxford Nanopore's chief technology officer.

But Oxford Nanopore will face stiff competition. Jonathan Rothberg, a scientist and entrepreneur who founded rival firm 454 Life Sciences, also announced at the AGBT conference that his start-up company, Ion Torrent, will be launching a desktop sequencing machine. Dubbed the Ion Proton, it identifies bases by using transistors to detect hydrogen ions as they are given off during the polymerisation of DNA.

This device will be capable of sequencing a human genome in 2 hours for around $1000, Rothberg claims. Nanopores are an "elegant" technology, he says, but Ion Torrent already has a foot in the door. "As we saw last summer with the E. coli outbreak in Germany, people are already now using it," he says.

Pocketful of DNA

By contrast, the MinION would take about 6 hours to complete a human genome, Brown claims, though the company plans to market the device for use in shorter sequencing tasks like identifying pathogens, or screening for genetic mutations that can increase risk of certain diseases. Each unit is expected to cost $900 when it goes on sale later this year.

"The biggest strength of nanopore sequencing is that it generates very long reads, which has been a limitation for most other technologies," says Loman. If the costs, quality, ease of use and throughput can be brought in line with other instruments, it will be a "killer technology" for sequencing, he says.

As for clinical applications, David Rasko at the Institute for Genome Sciences at the University of Maryland in Baltimore, says the MinION could have huge benefits. "It may have serious implications for public health and it could really change the way we do medicine," he says. "You can see every physician walking around the hospital with a pocketful of these things." And it will likely increase the number of scientists generating sequencing data by making the technology cheaper and more accessible, he says.

Beloved S.African musician, 60, back at school (AP)

JOHANNESBURG ? The 60-year-old beloved South African musician shows up five days a week to sit behind a scarred desk in a bare-bones classroom, studying for the high school diploma he missed when he dropped out to pursue a career in rock, funk, soul and jazz.

And Sipho "Hotstix" Mabuse wants the faith he is showing in learning to inspire a battered community.

Returning to school "is for me. But it's not about me," Mabuse said in an interview in his Soweto home, decorated with souvenirs from his European, U.S. and Mideast travels.

Mabuse, his round, unlined face animated beneath a shaven pate, said some of his friends and fellow musicians were baffled when he told them he was going back to school for what's known here as a National Senior Certificate, or, more commonly, a matric.

But bassist and producer Victor Masondo wasn't surprised.

"Sipho is a perfectionist," Masondo said. "In typical Sipho style, he wants to go through the right routes and do the right thing."

Mabuse often paces his tile floors as he practices his saxophone. He also plays the flute, piano and other instruments as well as the drums that gave him his nickname.

He has performed with Percy Sledge, Wilson Pickett, Miriam Makeba, Hugh Masekela and Paul Simon, giving the last a tour of Soweto when the American was here to work on "Graceland."

Mabuse also has sat on the boards of South Africa's National Arts Council and the Southern African Music Rights Organization, which works to protect musicians' intellectual property rights.

"He's always been there for South African music," said South African composer and theater director Welcome Msomi.

With his money, Mabuse could have hired a tutor to get him through the matric exams. With his connections, he could have found a university to take him on as a researcher. Instead, he's enrolled in classes offered to adults after the day of learning for younger students is complete. The public school is just around the corner from his home in Soweto, the township where he was born and raised.

Japie Masombuka, who runs the adult education program at Thaba-Jabula Secondary School, said he asked his most famous student to give a motivational talk one afternoon. To Masombuka's surprise, even students who were habitually late to class or often skipped class showed up, and now many are more responsible.

One of Mabuse's classmates, 27-year-old Nikiwe Mpande, left high school to take a secretarial course and start work. She said the lack of a matric was keeping her from getting promotions, but she didn't seriously consider returning to school until she heard about Mabuse.

He's shown her "you're never too old to actually finish school."

Mabuse is a local hero in Soweto, known as a cauldron of resistance to apartheid. In 1976, Soweto high school students set off nationwide protests when they rose up against inferior education.

Apartheid politicians created curricula meant to ensure blacks could aspire to be no more than servants for whites. For decades, the bulk of spending on education was reserved for the white minority.

Apartheid ended in 1994, but most teachers today are the products of apartheid education. They are ill-equipped to prepare their students for the 21st century, which may explain some of the angry frustration that characterizes frequent teacher strikes for higher pay. The students, taking a cue from their teachers, treat their schools with contempt, arriving late and showing little discipline in class.

Across South Africa, more than half a million members of the class of 2011 dropped out or otherwise disappeared before the 496,000 remaining took their final exams. Only a quarter of those who graduated did well enough to qualify for university study, according to government figures.

A government report released last month said public adult education programs haven't escaped the chaos. According to the report, facilities are spare, curriculum unfocused, teachers weak, and most of the more than 300,000 students who enrolled in such classes along with Mabuse last year failed to progress.

In the 1960s, Mabuse's Orlando West High School had no library or science lab. Many schools in impoverished black communities still lack such basics.

But Mabuse said Orlando West did have a respected principal, Solomon Kgokgophana Matseke, who died last year. Mabuse keeps a scrap book on Matseke on his bookshelf alongside biographies of post-apartheid South Africa's first two presidents, Nelson Mandela and Thabo Mbeki.

Matseke, himself a musician and composer, tried to persuade Mabuse to stay in school back in 1969, Mabuse said. But he and the fellow classmates who had formed a band were making money and earning admiration from girls at weekend performances.

One day, his mother went looking for Mabuse at Orlando West and no one could tell her where he was. She found him and his bandmates at a tailor's, trying on new stage costumes.

"She just said, `Oh, so this is your new school uniform?'" Mabuse recalled, saying she then dragged him back to school, where a teacher administered a beating.

When Mabuse has to miss class these days, he calls his teachers to apologize. He addresses teachers who are 20 years his junior as sir. And while the teachers might keep quiet when fellow students in their 20s answer their cell phones in class, Mabuse is not shy.

"I do try to instill that level of responsibility," he said. "I would chide some of them, `It's either you behave like school kids, or you behave like adults.'"

Mabuse also has been known to scold children he runs into in the streets during the day, telling them they should be in school.

Post-apartheid South Africa, Mabuse said, needs an educated public to protect its fragile democracy.

"On the continent, presidents are like kings. We cannot challenge them, because we have little knowledge," he said.

Mabuse's own pursuit of knowledge hit a setback last year, when he tried to take all six classes he needed to obtain his matric. He was too impatient to listen to teachers who told him that was too heavy a load for a man with his busy schedule. He failed two classes.

This year, he's confident he'll pass his remaining classes. And he says he's not embarrassed to tell people he had to try again.

He plans to go on to university to study musical anthropology. His goal is to write a book after interviewing his elders about what has shaped traditional music in South Africa, and how those traditions still ring.

He grew up listening to his grandfather and uncles singing a traditional choral form known as scatamiya.

"Maybe I can make a contribution in how we see ourselves. How we hear ourselves," Mabuse said.

    AP Interview: Homecoming inspires wounded Marine (AP)

    JACKSONVILLE, Ala. ? Shot through the upper back on a rooftop in Afghanistan and gasping for breath after a bullet ripped through his chest, Marine Sgt. Ben Tomlinson had a fear worse than death.

    Heat and shock from the round damaged his spinal cord, paralyzing him from the chest down. This injury was so much worse than one nine months before, when he'd been hit by a mortar blast during his first tour in Afghanistan. Yet through all the pain, Tomlinson was haunted by the idea people back home wouldn't remember him, that they wouldn't even know he was hurt.

    "That's the worst fear for everyone that gets deployed," said Tomlinson, who now uses a wheelchair. "It's that no one is thinking about them."

    Tomlinson's hometown of Jacksonville, in northeast Alabama, made sure he wouldn't feel forgotten, staging a welcome home celebration veteran groups say should be a national model now that fighting has ended in Iraq. While St. Louis is the only large city to hold a major parade for returning veterans, thousands of people lined the streets of Jacksonville and packed the town square to greet Tomlinson upon his return from the war, waving flags and giving money to help make his home more wheelchair accessible.

    "We need this to happen much more around the country, but (I) do not believe it actually occurs as much as it is needed because the wars have been going for such a long period of time," said Vietnam veteran John Stewart, who heads Operation Welcome Home in Inverness, Fla.

    More than being just a one-day morale boost, Tomlinson said the lavish homecoming in the city of 7,700 people inspired him to work harder on his rehabilitation so friends and neighbors knew he is trying hard to get better.

    "This is not the way I am going to be forever," said Tomlinson, 24, working out with dumbbells in the TV room at his home. He's getting stronger little by little, his biceps filling out after months shuttling between hospitals and doctor's appointments, and he's hopeful of walking again one day.

    Tomlinson played football and baseball and ran track at Jacksonville High School before graduating in 2006 and going to Mississippi College to play football. But college ball just wasn't the same as high school athletics, and he took a buddy's suggestion to consider the Marines. He was interested in joining a Force Reconnaissance squad, the "baddest of the bad," as Tomlinson put it. These are the Marines on the tip of the spear, the troops who go first into hostile areas.

    "It sounded pretty intense," said Tomlinson. It was.

    Tomlinson arrived in Afghanistan in May 2009. Three months later, while securing a road during a drug raid in a marketplace, a mortar hit the top of a building near where he was standing. Shrapnel peppered his body.

    "I got a piece in the neck, a piece hit me in the nose, but the worst part of that was getting a piece stuck in the radial nerve in my arm," said Tomlinson. "There's really no relief from pain when you have something stuck in your nerve."

    Surgery followed, and Tomlinson was back on duty three weeks later after taking a cast off his arm with the help of a medic, who removed his stitches. Tomlinson's tour of duty ended but he was back in Afghanistan for another seven-month stint by November 2010.

    With only one or two missions left on his second tour, Tomlinson was atop a roof in an Afghan village controlled by the Taliban while other troops looked for weapons and bomb-making materials.

    "We were doing that for about five or six hours. I want to say it was the middle of the day, and I got up to move and as soon as I got up ...," Tomlinson said. "It wasn't like I heard anything or really felt anything. It was like a sudden shock to my body, and then my vision went kind of obscured, and I couldn't move and I just started falling back and `boom,' hit the roof, and that's when I realized I'd probably been hit."

    The slug entered the back of Tomlinson's left shoulder near the base of his neck and traveled to his chest, damaging his spine and leaving him unable to move from the chest down. Evacuated from Afghanistan first to Germany and then the United States, he spent months at a hospital in Tampa, Fla., before returning home to Alabama on Jan. 18 to a hero's welcome: People lined the roads for miles leading from the airport to Jacksonville, and U.S. flags were everywhere. Businesses all over town put out welcome home signs.

    Several thousand people filled the town square for a ceremony, and some 800 students and faculty from Jacksonville High greeted him at his house, which is directly across the street from the school.

    "All I could say was, `Wow,'" Tomlinson said.

    With such a welcome home, Tomlinson said he knew he hadn't been forgotten ? his worst fears weren't realized.

    "People I didn't even know were thinking about me when this happened," he said.

    Stewart, himself a Vietnam veteran, said such receptions can only help troops coming home from multiple deployments after a decade of fighting in Iraq and Afghanistan.

    "Everyone is familiar with the horrendous reception we Vietnam veterans received when returning from the war and we must never, ever let that happen again," he said. "We must honor our heroes. We must remember their sacrifices."
